Protective Effects of Crocin on Experimental Gastrocnemius Muscle Ischemia/Reperfusion Model in Rat

The aim of the present study was to define whetherCrocin, has a regulatory effect againstexperimental gastrocnemius muscleischemia/reperfusion injury model in rats.A total of 32 Sprague-Dawley male,200-250g rats were randomly divided into four groups: Control, Crocin(60mg/kg/i.p), I/R and I/R+Crocin groups. All rats were anesthetized withxylazine and ketamine. Except for the control and Crocin group, the left lowerlimbs of the other 2 groups were applied to 2 hours of ischemia and 2 hours ofreperfusion with tourniquet. SOD, MDA, GSH, CAT, GPx, MPO, NO, IMA, AOPP ve PONlevels were measured in the plasma; the levels of SOD, MDA, GSH, CAT and GPxwere determined in gastrocnemius muscle. Increases in SOD, GSH, CAT, GPx, AOPPand PON levels, decreases in the MDA, MPO, NO and IMA levels were significantin the Crocin groups according to the I/R group. These differences werestatistically significant. The results of this study support the possibilitythat Crocin may play a protective role against skeletal muscle injury caused byI/R in rats by reducing oxidative stress. Crocin can be safely used forischemia/reperfusion injury.Downloads
